Objective To investigate the effects of eleutheroside E (EE) on bone mass and neuropeptides in ovariectomized (OVX) mice. Methods Twenty-four 7-week-old C57BL6/ J female mice were randomly divided into Sham group (Sham), OVX group, EE low-dose group ( EE-L), and EE high-dose group ( EE-H). On the basis of OVX, mice in EE group received 20 mg/ (kg· d) of EE-Land 40 mg/ ( kg· d ) of EE-H by gavage. Mice in Sham and OYX groups were gavaged with an equal volume of 1 % carboxymethyl cellulose Na. After 12 weeks, the left femur and serum samples were collected. Micro-CT scan was performed on the left femur. The contents of TRAP, PlNP, TNF-a, IL-6, IL-10, CORP, SP, VIP, and NPY in serum were detected using ELISA. Results Compared with those in Sham, the levels of BMD, BV /TV, Tb.N, PlNP, IL-10, CORP, SP, VIP, and NPY decreased significantly in OVX group ( P<0. 05, P<0. 01), while Tb.Sp, TRAP, TNF-a and IL-6 increased significantly in OVX group ( P<0. 05). Compared with those in OVX, the levels of Tb.N, VIP and NPY in EE-L increased significantly ( P< 0. 05). BMD, BV /TV, IL- IO, CORP, and SP showed upward trend, but there was no significant difference ( P>0. 05). Tb. Sp, TRAP, TNF-a, and IL-6 showed a declined trend, but there was no significant difference ( P>0. 05). There was no change trend in Pl NP (P>0. 05). In EE-H group, BMD, BV/ TV, TB.N, PlNP, IL-10, CORP, SP, VIP, and NPY increased significantly (P<0. 05, P<0. 01), while Tb. Sp, TRAP, TNF-a, and IL-6 decreased significantly ( P < 0. 05). Conclusion EE significantly increases bone mass after OVX, possibly by reducing inflammation and increasing the content of neuropeptides in a dose-dependent manner. [ABSTRACT FROM AUTHOR]
目的 人饼究刺五加苷E ( Eleutheroside E, BE) 对去卵梨(OVX)小鼠骨量以及神经肽的影响。方法 ”选反24 只7周龄 C57BL6/J 雌性小鼠, 平均分为假手术组(Sham 组) .OVX 组.EE 低剂量组 (EE-L 组)和EE 高剂量组(EE-H 组), EE-L 组和 EE-H 组在 OVX 基础上分别予 EE 20 mg/(kg . d)和40 mg/(kg . d)灌胃,Sham 组和OVX组以等体积 1%羟申基纤维素钠 (CMC-Na)灌胃。12 周后收集小鼠左侧股骨和血清标本。左侧股骨行 Micero-CT 扫撒;ELISA 法检测血清中骨上吸收指标抗酒石 酸酸性磷酸酶(TRAP) .骨形成指标 I 型原胶原氨基端捧(PINP) .炎性细胞因子肿瘤坏死因子-α(TNF-α) .白细胞介素 6(IL- 6) `昌细胞介素 10(IL-10) 以及神经肽降钙素基因相关肽(CCRP), 神经肽物质(SP) 血管活性肠肽(YIP)》、神经肽YY (NPY)的含量。结果 与 Sham 组相比,OVX 组小忌骨密度 (bone mineral density, BMD ). 骨体积分数(BVZTV).骨小梁数量 (Tb.N) .PINP IL-10.CGRP SP VIP 和 NPY 昌著性降低 (P<0.05, P<0.01), 骨小梁分离度(Tb.Sp) .TRAP TNF-a 和 II-6 昌 著性增高(P<0.05) ;与 OVX组相比,EE-L 组小鼠 Tb.N .VIP 和 NPY 显著性增高 (P<0.05), BMD 、BVZTV IL-10.CGRP 和 SP 有增加趋势, 但无显著性差异(P>0.05), Tb.SpTRAP TNF- 和工-6 有降低趋势, 但无显著性差异 (P>0.05), PINP 未见改变 趋势 (P>0.05); EE-H 组小鼠 BMD 、BVZTV Tb.N .PINP IL-10.CGRP、SP VIP 和NPY 显著性增高( P<0.05, P<0.01), Tb.Sp、 TRAP TNF-α 和IL-6显著性降低(P<0.05)。结论 EE 能够显著增加 OVX 骨量, 其机制可能是通过降低炎症反应, 增加神经 肽的含量而实现,并呈现剂其依赖性。 [ABSTRACT FROM AUTHOR]
