苗药花蝴蝶回调CIA 模型大鼠脂肪细胞变化的研究.

Objective To investigate the retracement on related indexes and mechanism of adipocytes changes in CIA model rats with Miao Polygonum runcinatum B.-H.v.S H. Methods A total of 40 rats were randomly divided into normal group. CIA model group methotrexate group. Polygonum runcinatum B.-H. v. S H. high dose group and low dose group. respectively. With 8 rats in each group. According to the relevant literature and the experience of folk medicine, intragastric administration was given respectively. (normal group: normal saline: methotrexate group: 1. 04mg/kg methotrexate. twice a week: Polygonum runcinatum: B.-H. v. S H. high dose group and low dose group: 7. 6.3.8 mg/kg Polygonum runcinatum B. H. v. S H. liquid per day), and stopped after 35 days. The foot swelling of rats in each group was observed, and blood triglyceride (TG), rheumatoid factor (RF). immunoglobulin A (IgA), IgG, IgM and other indicators were observed. Enzyme-linked immunosorbent assay was used to detect adiponectin (ADPN), interleukin-10 (IL-10) and other cytokines were detected and pathological sections of foot joints were studied. The experimental data were statistically processed by SPSS25, 0 software. Results The joint swelling index score and RF-IgM levels in the CIA model group were significantly higher than those in the other groups. IgG level was slightly lower than the Polygonum runcinatum B.-H. v. S H. low dose group and higher than other groups and the differences were statistically significant (P<0.05). The levels of TG. ADPN and IL-10 were significantly lower than those in the other groups. and the differences were statistically significant (P<0,05). In the CIA model group. there were low-grade adipocytes in the bone marrow cavity of the foot joints of the rats and the white blood cells were diffusely proliferated. Compared with other groups. the differences were statistically significant (P<0.05). Conclusion Miao Polygonum runcinatum B.-H. v. S H. may ensure normal energy metabolism (such as TG. etc.) and immunomodulatory factor secretion (such as ADPN. IL-10, etc.) by reversing the reduction of adipocytes in CIA model rats, promote immunomodulatory effects (such as reversing RF. 目的 探讨苗药花蝴蝶回调牛二型胶原蛋白诱导关节炎(CIA)模型鼠脂肪细胞变化的相关指 标及其机制。方法 将40只雄性SD大鼠随机分为正常组(8只)及建模组(32只),建模组大鼠建模成功后分 为 CIA 模型组、氨甲蝶呤组及花蝴蝶高剂量组、低剂量组,各8只,根据相关文献资料结合民间用药经验分别灌 胃给药(正常组:生理盐水;氨甲蝶呤组:1.04mg/kg氨甲蝶呤,每周2次;花蝴蝶高、低组:每天7.6、3.8mg/kg 花蝴蝶药液),35d后停止。观察各组大鼠足肿胀情况,检测血液甘油三酯(TG)、类风湿因子(RF)、免疫球蛋白 A(IgA)、IgG、IgM 等指标,采用酶联免疫吸附测定法检测脂联素(ADPN)、白细胞介素-10(IL-10)等细胞因子, 研究足关节病理切片等。采用SPSS25.0软件对实验数据进行统计学处理。结果 CIA 模型组大鼠关节肿胀 指数评分及 RF、IgM 水平均明显高于其他组,IgG 水平低于花蝴蝶低组而高于其他组,差异均有统计学意义 (P<0.05),TG、ADPN、IL-10水平明显低于其他组,差异均有统计学意义(P<0.05)。CIA 模型组大鼠足关 节骨髓腔内现低度脂肪细胞,而白细胞弥漫性增生,与其他组比较,差异均有统计学意义(P<0.05)。结论 苗 药花蝴蝶可能通过回调 CIA 模型鼠脂肪细胞的减少,保证了正常的能量代谢(如 TG 等)和免疫调节因子分泌 (如 ADPN、IL-10等),促进了免疫调节作用(如回调了 RF、IgG、IgM 等),缓解了足关节炎症等。 [ABSTRACT FROM AUTHOR]
