The Effect of Picloram and Thidiazuron Concentrations on Proliferation Somatic Embryos from Immature Inflorescence of Date palm. (English)

Immature inflorescences of date palm (Phoenix dactylifer) cv. Sewi were used as explants for in vitro culture to investigate the effect of plant growth regulators on inflorescence proliferation. The inflorescences on initiation stage cul-tured on solidified Murashige and Skoog (MS) basal medium supplemented with Picloram 1, 5, 10 and 15 mg l -1 for two re-cultures. Then, transferred into me-dium supplemented with different concentrations of TDZ combined with NAA on proliferate stage. The optimal concentrations for successful inflorescence growth was 5 or 10 mg l -1 Picloram and through studying the residuals effect of Picloram on inflorescences proliferation in the presence of three concentrations of TDZ, it found that, 0.5 mg l -1 TDZ combined with 0.1 mg l -1 NAA was more effective to induce direct somatic embryos and gave the highest inflorescence proliferation percentage, while the high level of Picloram induced callus. Vegeta-tive shoots formed into media containing 0.1 NAA mg l -1 , 0.2 mg l -1 BA and 0.2 mg l -1 kin. All somatic embryos were converted successfully to healthy normal plantlets which could be transferred to greenhouse. [ABSTRACT FROM AUTHOR]

اجرى هذا البحث لدراسه تاثير منظمات النمو على تخليق النورات الزهريه غيرالناضجه لنخيل البلح . تم زراعه النورات الزهريه فى مرحله البدايه على بيئه موراشيجى وسكوج مزوده بتركيزات من البيكلورام (1،10,5،15 مللجرام/ لتر) لمده نقلتين , اما فى مرحله التكشف فيتم زراعتها على بيئه محتويه على تركيزات من الثيديازورن مع تركيز واحد من النفثالين اسيتك اسيد. لوحظ ان افضل تركيز اعطى نمو للبراعم كان 5 او 10 مللجرام/ لتر بيكلورام . وعند دراسه التاثير المتبقى لماده البيكلورام على تكشف البراعم الزهريه فى وجود ثلاث تركيزات من الثيديازورن والنفثالين اسيتك اسيد. وجد ان 0.5 مللجرام/ لتر الثيديازورن مع 0.1 مللجرام/ لتر نفثالين اسيتك اسيد اكثر فاعليه لاحداث اجنه جسميه مباشره وكذلك تعطى افضل نسبه تكشف للاجنه المباشره , بينما تؤدى التركيزات العاليه من البيكلورام الى تكوين كالس. يتم تكوين النموات الخضريه على بيئه محتويه على 0.1 مللجرام/ لتر نفثالين اسيتك اسيد , 0.2 مللجرام/ لتر كينتين وكذلك 0.2 مللجرام/ لتر بنزيل ادنين . وقد تحولت الاجنه الجسميه بصورة طبيعية إلى نبيتات جيدة النمو يمكن أقلمتها بنجاح.
