DENEYSEL MULTIPL SKLEROZ (MS) FARE MODELİNDE SCHİSANDRİN B VE ALANTOLAKTON'UN İMMÜNOMODÜLATÖR ETKİLERİNİN ARAŞTIRILMASI. (Turkish)

Multiple sclerosis (MS) is a chronic inflammatory demyelinating disease that affects the central nervous system. MS is a chronic disease in especially young adults characterized by findings affecting the spinal cord white matter in a multiple localization. It begins between the ages of 20-40 and affects around 2.5 million people world wide. MS is a disease that reduces the quality of life with physical and psychological damages caused in the young in dividuals and imposes economic burdens on the patient / health system. Many anti-inflammatory, immuno modulatory and immuno suppressive drugs used in the treatment of MS reduce relapses but show limited effects in progressive disease. In this sense, it is necessary to discover new agents that can show axon-protective approach to treat or prevent MS. In this study, in experimental autoimmune encephalomyelitis (EAE) mouse model, by the purpose to treatment, respectively; The active substances of the most abundant Schisandrin B (Sch-B) in the Schisandra chinensis plant and Alantolactone (ALT) which a sesquiterpene lactone isolated from Inulahelenium, were used. Studies with these active ingredients have been shown to affect adjuvant T celse it her directly or indirectly. In this study, Sch-B and ALT significantly reduced EAE clinical scores. At the same time, these two molecule is also reduced demyelination. Cells from the brain, spinal cord, spleen and lymphnode were analyzed by flowcytometry, and these two molecule shave been shown to affect CD4 + T cells. In the light of the information obtained from our results, Sch-B ve ALT are could thought to be effective drug candidates in the prevention of MS. [ABSTRACT FROM AUTHOR]
Multipl skleroz (MS), merkezi sinir sistemini etkileyen, kronik inflamatuardemiyelinizan hastalığ ıdır. O zellikle MS ğenç erişkinlerde ğo ru len, omurilik beyaz cevherini birçok lokalizasyonda etkileyen bulğularla karakterize olan kronik bir hastalıktır. MS 20-40 yaş arasında başlar ve du nya ğenelinde yaklaşık 2.5 milyon kişiyi etkilemektedir. MS ğenç bireylerde neden olduğ u fiziksel ve psikolojik hasarlarla yaşam kalitesini du şu ren ve hastaya/ sağ lık sistemine ekonomik ku lfetler yu kleyen bir hastalıktır. MS tedavisinde kullanılan birçok antiinflamatuar, immu nomodu lato r ve immu nosu presif ilaç, relapsları azaltmakta fakat proğresif hastalıkta sınırlı etki ğo stermektedir. Bu anlamda MS'i tedavi etmek ya da o nlemek için akson koruyucu yaklaşım ğo sterebilecek yeni ajanların keşfedilmesi ğerekmektedir. Bu çalışmada, Deneysel Otoimmu n Ensefalomiyelit (EAE) fare modelinde tedavi amacıyla sırasıyla; Schisandrachinensis bitkisinde en bol bulunan Schisandrin B (Sch-B) ve Inulahelenium'dan izole edilen bir seskiterpenlakton olan Alantolakton (ALT) etken maddeleri kullanılmıştır. Bu etken maddelerin, yapılan çalışmalarda yardımcı T hu crelerini doğ rudan ya da indirek yollarla etkilediğ ini ğo sterilmiştir. Bu çalışmada, Sch-B ve ALT, EAE klinik skorunu anlamlı du zeyde du şu rmu ştu r. Aynı zamanda bu iki moleku l demiyelinizasyonu da azalmıştır. Beyin, spinalkord, dalak ve lenf nodundan alınan hu creler akım sitometri ile analiz edilmiş ve bu iki moleku lu n CD4+ T hu crelerini etkilediğ i ğo sterilmiştir. Sonuçlarımızdan elde edilen bilğilerin ışığ ında, Sch-B ve ALT'nin, MS'in o nlenmesinde etkili ilaç adayları olabileceğ i du şu nu lmu ştu r. [ABSTRACT FROM AUTHOR]
