Electron microscopic analyses of doublecortin, A neurogenesis marker, and gamma-aminobutyric acid immunoreactivities in wistar and genetic absence epilepsy rats from strasbourg

Amaç: Genetik absans epilepsili sıçanlarda (GAERS) hipokampal nörogenez araştırılmamış bir konudur. Çalışmamızda 21 günlük ve 3 aylık erişkin Wistar ve GAERS sıçanlarda nörogenezi karşılaştırmak ve yeni oluşan nöronların GABAerjik terminallerle sinaps yapıp yapmadığını incelemek amaçlanmıştır.Gereç ve Yöntem: Wistar ve GAERS (21 günlük ve 3 aylık) sıçanlardan perfüzyon fiksasyonu ile elde edilen beyin dokuları elektron mikroskopik analizler için hazırlandı. Hipokampal dentat girus (DG) bölgelerinden alınan ince kesitler immün-altın yöntemi ile anti-gama-aminobütirik asit (GABA) ve anti-doublecortin (DCX) antikorlarıyla ikili olarak işaretlendi. Elektron mikroskobunda incelenen kesitler morfolojik ve niceliksel analiz için fotoğraflandı. Bulgular: Tüm gruplarda DCX immünreaktivitesi akson, dendrit ve hücre gövdelerinde görüldü. DCX (+) yapıların GABAerjik terminaller ile sinaps ve sinaps benzeri yapılar oluşturduğu belirlendi. Tüm deney gruplarında, aksonlara kıyasla dendritlerdeki ve simetrik sinaps oluşturan yapılara kıyasla da asimetrik sinaps oluşturan yapılardaki DCX işaretlenmesinin istatistiksel olarak anlamlı ölçüde fazla olduğu belirlendi. Kalitatif gözlemlerde 21 günlük hayvanlarda 3 aylıklara kıyasla DCX (+) mossy terminallerinde (MT) daha fazla GABA olduğu görüldü. GABA içeren DCX (+) profiller 21 günlük gruplarda 3 aylık gruplara göre artış eğilimi gösterdi ancak istatistiksel anlamlılık saptanmadı. Ayrıca 21 günlük ve 3 aylık GAERS gruplarında istatistiksel anlamlılık olmasa da GABA ve DCX yoğunluklarında kontrollere göre artma eğilimi gözlendi. Sonuçlar: Elde ettiğimiz veriler yeni oluşan DCX-ir nöronların lokal hipokampal ağa katıldığını göstermektedir. 21 günlük hayvanlarda DCX (+) profiller 3 aylıklara kıyasla daha fazla GABAerjik olma eğilimindedirler; bu da immatür hayvanlarda GABA'nın trofik etkisinden kaynaklanıyor olabilir. İmmatür hayvanlarda DCX (+) MT'lerde daha fazla GABA görmemiz, postnatal gelişimin erken dönemlerinde MT'lerden GABA salındığına dair kanıtları desteklemektedir. Çalışmamızda GAERS gruplarında DCX ve GABA yoğunlukları artma eğilimi göstermiştir. Bu bulgular absans epilepsi mekanizmalarında hipokampustaki değişikliklerin de etkili olabileceğini düşündürmektedir.Anahtar Sözcükler: GAERS, nörogenez, doublecortin, GABA, elektron mikroskopi. Aim: Hippocampal neurogenesis is an issue which has not been investigated in genetic absence epilepsy rats from Strasbourg (GAERS). In the present study, we aimed to investigate neurogenesis in 21-day- and 3-month-old adult GAERS strain to determine if newly formed neurons form synapses with GABAergic terminals and compare the results to the control Wistar rats. Materials and Methods: Brain tissues obtained from all experimental groups by perfusion fixation were processed for the electron microscopic assessments. Thin sections from hippocampal dentate gyrus (DG) regions were double-labeled for anti-gamma-aminobutyric acid (GABA) and anti-doublecortin (DCX) antibodies using immunogold method. Sections, examined with the transmission electron microscope, were photographed for the morphological and quantitative analyses. Results: DCX immunoreactivity was demonstrated on the axons, dendrites and cell bodies in both Wistar rats and GAERS. We observed that DCX (+) structures formed synapses and synapse-like regions with GABAergic structures. Dendrites compared to axons, and profiles forming asymmetrical synapses compared to those forming symmetrical synapses had significantly increased DCX labeling in all experimental groups. In qualitative observations, 21-day-old animals were seen to have more GABA in their DCX (+) mossy terminals (MT), compared to 3-month-old animals. DCX (+) profiles including GABA showed a tendency to increase in 21-day-old groups when compared to 3-month-old groups, although no statistical significance was determined. Besides, there was a trend for increase in GABA and DCX densities in 21-day-old and 3-month-old GAERS groups compared to their Wistar controls, but the difference was not statistically significant. Conclusion: Our data demonstrated that newly born DCX-ir neurons join the local hippocampal network. In 21-day-old animals, DCX (+) profiles had an increased tendency to be GABAergic, compared to 3-month-old groups; this may result from the trophic effects of GABA in immature animals. Our findings showing more GABA in MTs of immature animals supports the evidence that GABA is released from the MTs at early stages during postnatal development. In our study, DCX and GABA densities had a tendency to increase in GAERS groups. The present findings suggest that the alterations in the hippocampus may also have effect on the mechanisms of absence epilepsy.Key words: GAERS, neurogenesis, doublecortin, GABA, electron microscopy. 114
