The existence of circulating endothelial cells (CECs), in periphery has been recently proved. The appearance of CECs in cancer patients is related with angiogenesis and tumour metastasis. In particular, an increase is observed in the number of CECs in cancer patients which leads to angiogenesis and it is believed that circulating progenitors endothelial cells (CEPs) take part in these procedures. The purpose of this study was to detect CECs in blood by rare event cytometry analysis in breast cancer patients, before and after vascular endothelial growth factor (VEGF) antibody blockade by bevacizumab (Avastin) infusion and in prostate cancer patients before and after endoxan infusion. The peripheral blood of 11 breast cancer patients and 4 control normal aged and sex matched samples and 4 prostate cancer patients and 2 control samples, was used. Samples were enriched with CD146+ cells, using immuno-magnetic separation from whole blood, and then CECs were standardized and counted by triple color flow cytometry analysis. In 6 out of 11 patients the number of CECs was decreased after bevacizumab infusion. In 2 patients there was an increase of CECs after Avastin infusion, while in 1 there was no significant change in the number of CECs and 2 did not attend the follow up. In prostate cancer patients, the number of CECs was high before treatment and it seems that it is decreased after endoxan infusion. Understanding the function of CECs, may help in their characterization as biological prognostic and follow-up markers of patients with metastatic tumors. Furthermore, it is necessary to investigate the angiogenic ability of CEPs in these patients in conjunction with other factors such as VEGF.
