Brassicaceae family contains a high number of cultivated species that are economically important worldwide. However, the plants of this family are susceptible to many pathogens including viruses and phytoplasmas. The present thesis studied the incidence of Cauliflower mosaic virus (CaMV), Turnip mosaic virus (TuMV), Tomato spotted wilt virus (TSWV), Cucumber mosaic virus (CMV) and Lettuce mosaic virus (LMV) mainly in the three widely cultivated species of Brassica genus, Brassica oleracea var. capitata, Brassica oleracea var. botrytis and Brassica oleracea var. italica and also in arable weeds of various families. The samples of cultivated plants, collected from various regions of Greece and one region of Turkey, were examined by ELISA and the incidence of all five viruses was found in low rates. However, only the incidence of CaMV and TuMV was confirmed, with molecular methods (PCR) as well as with bioassays at indicator plants. Arable weeds from various regions of Greece were also examined by ELISA and the viruses were identified in a limited number of plants. However, virus presence has not been confirmed with mechanical inoculations or molecular methods. Finally, in Brassica plants exhibitng yellowing symptoms, Turnip yellows virus (TuYV) was also detected by RT-PCR. Some of the CaMV and TuMV isolates were partially characterized. TuMV isolates were found to belong in World-B and Basal-B groups whereas CaMV isolates were separated in four different groups based on the Restricted Fragment Length Polymorphism (RFLP) analysis using Hpy99I enzyme. The effect of mixed infection of CaMV and TuMV on the acquisition/transmission of each virus by Myzus persicae was also studied. The results showed that TuMV transmission efficiency was higher than CaMV. However, transmission of each virus from singly and doubly infected plants was similar. Finally, a phytoplasma belonging to the Aster Yellows (AY) group was detected in cabbage samples for the first time in Greece.
