Polihidroksibütiratlar (PHB) bakteriler ve arkeler tarafındanda sentezlenebilen makromoleküllerdir. Mikroorganizmalar olumsuz stres koşulları altında ürediklerinde PHB'ler inklüzyon cisimcikleri şeklinde karbon ve enerji kaynağı olarak hücre içerisinde birikirler. PHB'ler en iyi bilinen polihidroksialkonat (PHA)'lar olup Haloarcula, Haloferax ve Halorubrum sp. gibi bazı halofilik arkeler ve Bacillus sp. gibi bazı bakteriler tarafından üretilmektedir. Petrokimyasallardan elde edilen plastik maddeler doğada uzun yıllar çözünmediklerinden ciddi çevre sorunlarına neden olmaktadırlar. Doğal çevresel koşullardaki hızlı ve %100 çözünürlüklerinden dolayı PHB'ler biyolojik olarak çözünebilir plastiklerin üretimi için çok önemli bir alternatif sunmaktadırlar. PHB'ler polipropilen gibi çeşitli sentetik termoplastiklere benzer özelliklere sahip olan polimerler olduklarından çevre dostu biyoplastiklerin geniş kapsamlı uygulamaları ve gelecekteki ticari olarak seri üretilmesi için önemli adaylardır. Ancak, üretim maliyeti diğer plastiklerden daha yüksek olduğundan, etkin bakteri suşlarının geliştirilmesi, fermentasyon ve geri kazanım süreçlerinin optimizasyonu gibi çalışmalarla bu dezavantaj giderilmeye çalışılmaktadır. Bu nedenle, çalışmamızda daha önce Tuz Gölü'nden izole edilmiş olan 10 Haloarcula sp. suşları arasında rölatif %PHB birikimini karşılaştıralarak en iyi PHB üreticisi suşun tespit edilmesi amaçlanmıştır. Seçilen suştaki PHB üretimi kültür koşullarının (sıcaklık, pH, NaCl konsantrasyonu, inkübasyon süresi, besiyeri içeriği) optimizasyonu ile daha da arttırılarak biyoteknolojik uygulamalarda kullanılabilme potansiyelinin güçlendirilmesi sağlanmıştır. Polyhydroxybutyrates (PHB) are macromolecules synthesized by bacteria and archaea. When grown under different stress conditions, microorganisms accumulate PHBs inside the cell as carbon and energy source in the form of inclusion bodies. PHBs, the most well known polyhydroxyalkynate (PHA), are produced by some haloarchaea including Haloarcula, Haloferax and Halorubrum spp. and by some bacteria such as Bacillus sp. plastic materials obtained from petrochemicals cause serious environmental problems as they are not dissolved in nature. Due to their rapid and 100% solubility in natural environmental conditions, PHBs offer a very important alternative to the production of biodegradable plastics. Since PHBs are polymers with properties similar to synthetic thermoplastics such as polypropylene, they are important candidates for the wide range of applications of environmentally friendly bioplastics and their future commercial production. However, as production costs are higher than other plastics, efforts to improve effective bacterial strains, such as optimization of fermentation and recovery processes, are attempted to overcome this disadvantage. For this reason, in our study, relative PHB accumulation capacities of 10 Haloarcula sp. strains previously isolated from Lake Tuz, were aimed to compare to select the best PHB producer strain. The PHB production in the selected strain has been further enhanced by the optimization of culture conditions (temperature, pH, incubation time, nutrient content) with the aim of increasing its use in potential biotechnological applications. 82
