7-Cyano-7-deazaguanine is a biosynthetic precursor of the modified nucleotides of the 7-deazaguanosine-family as well as precursor to several natural products, such as sangivamycin and toyocamycin. The latter group of modified nucleotides and both sangivamycin and toyocamycin are characterized as widely distributed functional molecules of life and therapeutically-interesting compounds. The metabolic pathways of these molecules are extensively studied for many years but almost nothing is known about degradation pathways of the 7-deazaguanines. In this study we examined the degradation processes of 7-cyano-7-deazaguanine (PreQ0) in soil bacteria. From the soil samples we have isolated SINO2 bacterium that could transform the UV-Vis absorbance spectrum of the PreQ0. We have identified that SINO2 is capable to deaminate PreQ0 to 2,4-dioxo-2,3,4,7-tetrahydro-1H-pyrrolo[2,3-d]pyrimidine-5-carbonitrile, which might be the first biodegradation step of PreQ0. Based on 16s rRNA gene sequence, SINO2 could be attributed as the bacterium of the genus Microbacterium. By sequencing and analysing the genome of the bacterium, we chose three target deaminases CYDE1, CYDE2 and CMP deaminase. We have tried to synthesize these three recombinant deaminases and tried to perform PreQ0 deamination experiments with these enzymes. By modeling the structures of deaminases CYDE1, CYDE2 and CMP deaminase of Microbacterium sp. SINO2 bacterium and by performing protein-ligand docking experiments we speculate that CYDE2 may be the most feasible enzyme to catalyze the deamination of PreQ0 although this hypothesis should be tested experimentally. To the best of our knowledge, this is the first-time demonstration of how the 7-cyano-7-deazaguanine can be transformed to other compound while in biological setting.
