Surface plasmons (SPs) are the oscillation of electrons inside the metal longitudinally propagating through the interface between the metal and the dielectric matter, which are sensitive to the properties of both the metal and the dielectric matter. Changes in the properties of the dielectric matter will cause changes in both amplitude and phase of the reflected or transmitted light, which are used as signals for the detection of the dielectric matter. Phase sensitive Surface Plasmon Resonance (SPR) techniques are a popular means of characterizing biomolecular interactions, because of the characteristics of SPR techniques, e.g. label-free real-time measurement with high sensitivity. However, limitations due to the narrow dynamic range and difficulty in adapting the method for multi-point sensing have restricted its range of applications. This thesis presents a compact phase-sensitive SPR technology USIng a custom CMOS camera. The phase of p- and s- polarization is sinusoidally modulated by a phase modulator and demodulated by this custom CMOS camera. The signal adopted for the measurement of refractive index change on the sensor surface is modulation depth, which is the amplitude of the fundamental frequency divided by DC.
