Examine the interaction between Photensin and EHD Proteins

Eps15-homology domain containing 1 (EHD1) and EHD4 are the members of EHD protein family. Both are distinct from other EH family members with the EH-domain at the C-terminal region. EHD proteins play an important role in regulation of endocytic transport. Most of the EHD-binding proteins contain an Asp-Pro-Phe (NPF) motif that interacts with EH-domain of EHD proteins. Phostensin (Protein phosphatase 1 F-actin ctoskeleton targeting subunit), encoded by KIAA1949, consists of 165 amino acids, and contains a consensus PP1-binding motif and an actin-binding motif. GST pull down assay showed that EHD proteins, EHD1 and EHD4, directed bind to phostensin at the region between residues 65 and 80. Then we used full-length phostensin to pull down truncated EHD proteins, and showed that phostensin can bind with EHD proteins without EH doamin. Based on our study, we suggested that phostensin serves as a scaffold protein that associates with EHD1 (or EHD4), PP1 and actin, and modulates endocytic vesicle transport.
