A method of therapeutically treating a mammal having a cancerous tumor comprising cells that express a hedgehog polypeptide, wherein the method comprises administering to the mammal a therapeutically effective amount of a CDO antagonist that binds to the hedgehog polypeptide, thereby resulting in the effective treatment of the tumor.

A method of diagnosing the presence of a tumor in a mammal, comprising (a) contacting a test sample comprising tissue or cells obtained from the mammal with a molecule that binds to a CDO (cell adhesion molecule-related/down-regulated by oncogene) polypeptide; (b) detecting the formation of a complex between the CDO-binding molecule and the CDO polypeptide in the test sample, wherein the formation of more complex in the sample relative to a control sample is indicative of the presence of a tumor in the mammal.
  • Claim: 14. The method of claim 13, wherein the CDO-binding molecule employed is detectably labeled or attached to a solid support.
  • Claim: 15. The method of claim 13, wherein the test sample of tissue or cells is obtained from an individual suspected of having a cancerous tumor.

  • Claim: 29. The method of claim 13, wherein the molecule that binds to a CDO polypeptide is an anti-CDO antibody, a CDO-binding antibody fragment, a CDO-binding oligopeptide, or a CDO binding small organic molecule.
  • Claim: 30. The method of claim 29, wherein the molecule is an anti-CDO antibody or a CDO-binding antibody fragment.
  • Claim: 31. The method of claim 30, wherein immunohistochemistry or FACS (Fluorescence Activated Cell Sorting) analysis is used to detect the formation of a complex between the CDO-binding molecule and the CDO polypeptide in the test sample.
  • Claim: 32. The method of claim 30, wherein the anti-CDO antibody or CDO-binding antibody fragment are radiolabeled.
  • Claim: 33. A method of diagnosing the presence of a tumor in a mammal, comprising detecting the level of expression of a gene encoding a CDO (cell adhesion molecule-related/down-regulated by oncogene) polypeptide (a) in a test sample of tissue or cells obtained from said mammal, and (b) in a control sample of known normal non-cancerous cells of the same tissue origin or type; wherein a higher level of expression of the CDO gene, as compared to the control sample, is indicative of the presence of tumor in the mammal from which the test sample was obtained.
  • Claim: 34. The method of claim 33, wherein the level of expression of a gene encoding a CDO polypeptide is detected by means of fluorescent in situ hybridization (FISH), Southern blotting, Northern blotting, or polymerase chain reaction (PCR) techniques.
  • Claim: 35. The method of claim 34, wherein said PCR technique is real-time PCR.
  • Claim: 36. A method of determining the expression levels of CDO (cell adhesion molecule-related/down-regulated by oncogene) in a patient, comprising (a) administering to a patient a molecule that binds a CDO polypeptide, wherein said molecule is tagged with a detectable label, and (b) externally scanning the patient for localization of the label.
  • Claim: 37. The method of claim 36, wherein the CDO-binding molecule is an anti-CDO antibody, a CDO-binding antibody fragment, a CDO-binding oligopeptide, or a CDO binding small organic molecule.
  • Claim: 38. The method of claim 36, wherein the detectable label is a radioactive isotope or a fluorescent label.
  • Claim: 39. The method of any one of claim 13, 33 or 36, wherein the CDO polypeptide has a sequence that is at least 80% identical to the sequence of SEQ ID NO: 4.
