Thermal tolerance and stress responses of the ditch shrimp Palaemon varians under the combined effects of temperature and salinity

As global temperatures continue to rise due to global change, marine heatwaves (MHW) are also becoming more frequent and intense, impacting biodiversity. Organisms inhabiting shallow marine environments, such as the ditch shrimp Palaemon varians, are expected to be the most affected by rising temperatures. Thus, addressing species’ thermal ecology and climate extinction risk is crucial to foster climate -smart conservation of shallow water ecosystems. Here, we estimated upper thermal tolerance limits (Critical Thermal Maximum, CTmax) of P. varians and further tested the combined effects of salinity (20 and 40) and temperature (20 vs 23 or 26 ºC) in stress biomarker levels in the muscle of shrimps over a one -month exposure (day 7, 14, 21 and 28). Females displayed lower CTmax than males, which may have been associated to their larger size and higher energetic investment in offspring. Overall, smaller individuals were more resistant to higher temperature, a pattern in line with previous studies. The average CTmax value of P. varians recorded in the population studied was 37.3 °C, which is higher than CTmax values recorded for northern populations (UK 31 ºC and France 36 ºC). This clinal variation may indicate some level of local adaptation in tolerance limits. However, considering the maximum temperature recorded in the sampling site (35.5 °C), this species thermal safety margin is narrow at the study site (1.5 °C for females and 2.2°C for males). Multivariate analysis of biomarker data showed that temperature and exposure time had a significant effect on stress biomarkers, as opposed to salinity, with no major interactions being detected between factors (temperature, salinity, and time of exposure). The overall increase in biomarkers abundance and/or activity levels (glutathione-S-transferase - GST, catalase - CAT, lipid peroxidation - LPO, superoxide dismutase - SOD, Ubiquitin and heatshock protein 70 kDa -HSP-70) at elevated temperatures reveals the induction of oxidative stress and protein denaturation, as well as a fast and efficient cellular response from this species to counteract damage to cellular components. The negative correlation between individuals’ weight and HSP-70 expression also evidenced how this shrimp species can cope with thermal stress according to shrimp size. These findings indicate that the increase in frequency and duration of MHW may negatively impact this shrimp species. If sea surface temperature increasing rate continues, species from shallow marine environments may experience shifts in their physiological performance and population dynamics. Ultimately, metabolic changes will most likely occur, along with a potential prevalence of smaller-bodied organisms and a shift in sex ratio .
Há medida que a temperatura global continua a aumentar, devido às alterações climáticas, as ondas de calor marinhas (OCM) também se estão a tornar cada vez mais frequentes e intensas, impactando assim a biodiversidade. Os organismos que habitam ambientes marinhos pouco profundos, tal como o camarão das salinas Palaemon varians, são os que se antecipa que sofram mais com este aumento de temperatura. Assim, abordar a ecologia térmica das espécies e o seu risco de extinção é fulcral para desenvolver estratégias inteligentes de conservação dos ecossistemas marinhos pouco profundos. Neste estudo estimámos o limite máximo de tolerância térmica (Máximo Térmico Crítico, CTmax) da espécie P. varians e averiguámos os efeitos combinados da salinidade (20 e 40) e da temperatura (20, 23 e 26 °C) através dos níveis de biomarcadores de stress no músculo desta espécie, ao longo de um mês (dia 7, 14, 21 e 28) de exposição. As fêmeas revelaram ter um CTmax inferior ao dos machos, o que pode resultar do facto de estas apresentarem maiores dimensões e um investimento energético na reprodução superior. No geral, os indivíduos mais pequenos demonstraram ser mais resistentes a temperaturas mais elevadas, confirmando assim os resultados de estudos anteriores. A média do CTmax da população de P. varians estudada foi de 37.3 °C, sendo assim superior aos valores registados na europa do Norte (Reino Unido 31 °C e França 36 °C). Esta variação pode ser indicativa de adaptações locais associadas à tolerância térmica. No entanto, tendo em consideração a temperatura máxima registada no local de amostragem (35.5 °C) a margem de segurança térmica que esta espécie apresenta é bastante estreita (1.5 °C para fêmeas e 2.2 °C para machos). As análises multivariadas dos biomarcadores revelaram que a temperatura e o tempo de exposição têm impactos significativos nos níveis de expressão dos biomarcadores de stress, sendo que o mesmo não aconteceu em relação à salinidade e às diferentes interações entre os 3 fatores considerados (temperatura, salinidade e tempo de exposição). O aumento generalizado da abundância e/ou atividade dos biomarcadores (glutationa-S-transferase - GST, catalase - CAT, peroxidação lipídica - LPO, superóxido dismutase - SOD, Ubiquitina e proteína de choque térmico 70 kDa - HSP-70) em temperaturas elevadas revela a existência de stress oxidativo e de desnaturação proteica, tal como uma rápida e eficaz resposta celular por parte desta espécie na mitigação de potenciais efeitos deletérios. A correlação negativa registada entre o peso dos indivíduos e os níveis de expressão da HSP-70 também demonstrou que esta resposta ao stress térmico poderá depender do tamanho dos indivíduos. Os resultados desta investigação indicam que com o aumento e intensificação das OCM, a espécie de camarão P. varians, poderá vir a sofrer efeitos nefastos a nível celular. Se a taxa de aumento da temperatura continuar a aumentar, as espécies que ocupam ambientes marinhos pouco profundos poderão sofrer alterações na sua performance fisiológica e na dinâmica das suas populações. Deste modo, é espectável que se registem variações metabólicas, uma maior prevalência de organismos com dimensões mais reduzidas, e uma potencial alteração do rácio machos: fêmeas.
