Cysteine Functionalization in the Synthesis of Complex and Well-Defined Bioconjugates

Cancer is one of the leading causes of death worldwide and is a major economic burden for governments and health systems. To address this critical health problem and explore a major economic market, ample research has been performed in the development of novel therapies. Antibody-drug conjugates (ADCs) present themselves as clear front-runners in the battle against cancer. By merging the exceptional targeting ability of antibodies and the potency of powerful cytotoxic drugs, ADCs display high levels of selectivity, tolerability and cytotoxic activity. The success of an ADC is closely related with the careful optimization of its four major components: the antibody, the payload, the linker and the bioconjugation technology. The linker, in particular, must be stable in solution, and capable of releasing the payload upon a predetermined stimuli. Boronic acids have been shown to react with oxygen species (ROS) and were widely used as ROS-responsive handles. However, boronic acids display relatively low stability in physiological conditions and promiscuous reactivity with endogenous molecules, such as sugars and proteins. In this work we identified diazaborines as stable alternatives to boronic acids that retain traditional boronic acid reactivity. In our first work, we demonstrated that boronic acids could inhibit selectively Human Neutrophil Elastase (HNE) with low μM potency. HNE is a serine protease present in diverse inflammatory processes and often implicated in tumor progression and for which there is no know inhibitor marketed. After computer-assisted optimization of the scaffold we achieved molecules with IC50 2.7 ± 0.6 μM, which is similar to the activity of boronic acid analogues. Finally, the diazaborines were shown stable over 24 hours in different pHs and plasma, contrarily to boronic acids which displayed half-lives around two hours. After recognizing diazaborine’s stability, we hypothesized it could retain boronic acid’s reactivity towards ROS. Gratifyingly, they are swiftly oxidized in the presence of 10 equivalents of hydrogen peroxide and are able to release a molecular payload. After carefully studying the oxidation mechanism, we used diazaborines to generate bioconjugates with model peptides that were stable in buffer and responsive to hydrogen peroxide. Finally we designed a breast cancer targeting ADC by using a diazaborine linker to attach the cytotoxic drug SN-38 to an engineered trastuzumab. With this work we demonstrated that diazaborines are versatile molecules that could be very useful in designing new scaffolds for medicinal chemistry and chemical biology in the preparation of novel targeted drug conjugates.
O cancro é uma das causas de morte mais prevalentes em todo o mundo. Para além da elevada taxa de mortalidade e morbilidade associadas, o cancro representa também um encargo enorme para os governos e sistemas de saúde mundiais. Para resolver esta situação, a indústria farmacêutica, em conjunto com a comunidade académica, têm desenvolvido esforços intensos no sentido de desenvolver novas terapias. Das novas terapias, os conjugados fármaco-anticorpo (ADCs) têm vindo a destacar-se com uma das mais promissoras. Ao aliar a excecional seletividade dos anticorpos à enorme potência dos fármacos citotóxicos, os ADCs demonstram uma ótima seletividade, tolerabilidade e atividade citotóxica. O sucesso de um ADC está intimamente relacionado com a otimização racional de cada um dos seus quatro constituintes: o anticorpo, o fármaco, o espaçador químico e a tecnologia de bioconjugação. O espaçador, em particular, tem de ser estável em condições fisiológicas e, simultaneamente, ser capaz de libertar o fármaco quando reconhece certas condições predeterminadas. Os ácidos borónicos são moléculas que, devido à sua orbital p livre, possuem uma reatividade muito característica. Para além de complexarem com ligandos de oxigénio e azoto, os ácidos borónicos são capazes de reagir com espécies reativas de oxigénio (ROS), que estão normalmente sobre expressas no ambiente tumoral. Como tal, os ácidos borónicos têm sido amplamente usados como funcionalidades capazes de detetar e reagir com ROS. No entanto, os ácidos borónicos são relativamente instáveis em condições fisiológicas e têm uma reatividade promiscua com açucares e proteínas, o que origina numerosos problemas ao nível da sua farmacocinética. Como tal, para serem efetivamente usados em condições biológicas, eles têm sido encapsulados em estruturas supramoleculares que os protegem do meio fisiológico. Durante este projeto, identificamos as diazaborinas como alternativas estáveis aos ácidos borónicos mas que mantem a sua reatividade tradicional. Os ácidos borónicos têm sido recentemente utilizados no desenvolvimento de novos inibidores de proteases de serina mas o seu progresso tem sido temperado pelos seus problemas de farmacocinética. Na primeira parte do trabalho, demonstramos que as diazaborinas são capazes de inibir com elevada potência a Elastase dos Neutrófilos Humana (HNE), uma protease de serina presente em diversos processos inflamatórios e frequentemente associada à progressão de processos tumorais. O passo inicial de otimização da estrutura foi auxiliado por estudos computacionais que permitiram o desenvolvimento de uma molécula com um IC50 de 2.7 ± 0.6 μM. Este valor é comparável à atividade de ácidos borónicos com uma estrutura semelhante, demonstrando que as diazaborinas não perdem a reatividade clássica dos ácidos borónicos e mantêm a afinidade para proteases de serina.
